Effect of Impermeants on the Change of Weight in Cold-stored Rat Liver




Abstract
Anaerobic cold storage is a simple and efficient method for liver preservation. Recently University of Wisconsin(UW) solution was developed and human liver can be stored safely for up to 24 hours in UW solution. But liver preservation with UW
solution
exhibited a remarkable shrinkage of endothelial cell and hepatocyte which may be due to large amount of impermeants in UW solution.
So we studied the effect of impermeants on the change of weight in cold-stored rat liver with Eurocollins(EC) solution. UW solution, modiefied UW soluion(UW1: without hydroxyethyl starch, UW2; without h: droxyethyl starch, half amount of
lactobionate).
Twenty four Spargue-Dawley male rat were divided in four group, 6 rats each group. Cold solution was infused via aorta and portal vein. The livers were stored for 0, 6, 12, 24, 48hr, and weighed.
After 48 hours of storage, the livers stored in EC solution gained 21.9% of their weight and the livers stored in UW solution lost 17.0% of their weight. In UW1 solution, the livers also lost 16.8% of their weight after. In UW2 solution, however,
there
were little change of their weight during 48 hors.
In is suggested that UW solution causes hepatocyte dehydration and liver preservation could be further improoved by reducing the concentration of impermeants such as lactobionate in UW solution.
